With an Olis RSM 1000 spectrophotometer and GlobalWorks™ software, the wavelengths do not have to be known in advance. And one measurement, with one sample, is all that needs to be made.

Let's return to the original raw data set and move through the steps in the Olis software to fit them. From raw 3D file to fitted answer takes a few seconds.

Raw 3D file:
Collect kinetic data as a function of wavelength. With the Olis RSM 1000, each 2D scan is acquired in one millisecond (unless user-specified signal averaging during collection is requested). During a one-second collection period, 1,000 scans are collected.

Eigenvector display:
Hand all or part of the raw data to the Olis Global Fitting scheme. The first step is SVD, or factor analysis. Factor analysis finds things which are changing at some rate with some shape. The results are shown in "eigenvectors," graphical representations of shapes. The upper row shows scan eigenvectors; the lower row shows kinetic eigenvectors. Eigenvectors without structure are "noise." With these noise contributions isolated from the "information," the remaining noise is data-specific, not systematic.
